Acne is an inflammatory disease of the skin caused by excess sebum production contains free radical called Reactive Oxygen Species (ROS). The presence of ROS causes oxidative stress leading inflammation that could be main pathogenic factor of acne. Antioxidant has known as ROS scavenger that could be produced by plants such as Dadap serep (Erythrina subumbrans). Dadap serep is a traditional herbal medicine plant from Indonesia that known has ability to inhibit the growth of bacteria causing acne such as Staphylococcus epidermidis. However, the antioxidant activity of dadap serep leaves is not yet known, especially in an ability to prevent acne. This study aims to determine the potential of dadap serep leaves (E. subumbrans) as a natural source of antioxidants in preventing acne. This research was conducted by in silico using the PASS prediction, extraction of dadap serep leaves, macroscopic morphology analysis, and in vitro by antioxidant test using the DPPH (1.1-diphenyl-2picrylhydrazyl) with 70% ethanol as solvent. The result of this study showed that several flavonoids from dadap serep leaves such as 5-Hydroxysophoranone, Abyssinone V, and Eribraedin B have Pa value of more than 0.7 which indicates that the chances of finding these activities in laboratory tests are high based on PASS Prediction. Based on DPPH test, antioxidant activity of dadap serep leaves were very strong with an IC50 value of 8.32 ppm. Dadap serep leaves extract from Semarang, Central Java (Erythrina subumbrans) has potency to be antioxidant source to prevent acne.
