International Agency for Research on Cancer (IARC) has been classified acrylamide as “probably carcinogenic to humans” (group 2A). Acrylamide exposure in high concentration induced reactive oxygen species and generated 8-Hydroxy-2’-Deoxyguanosine (8-OHdG) leading to oxidative DNA damage. This study aimed to detect DNA Adduct 8-hydroxy-2’-deoxyguanosine (8-OHdG) as a biomarker oxidative DNA damage caused by reactive oxygen species from acrylamide exposure. This in vitro study was conducted by incubating 2-deoxyguanosine, one of the bases DNA, H2O2, and acrylamide as xenobiotic. The reaction was incubated at 37 °C for 24 hours and varied at pH 7,4 and 8,4. 8-Hydroxy-2’-Deoxyguanosine (8-OHdG) was analyzed by using reverse-phase HPLC with UV-Vis detector and an equipped mixture of phosphate buffer and methanol as mobile phase. The results showed that acrylamide could interact with DNA base, 2-deoxyguanosine, and induced 8-OHdG formation. The 8-OHdG concentration was higher from the exposure of 2-deoxyguanosine to acrylamide and H2O2 compared to the reaction without the addition of acrylamide. This result indicated that acrylamide exposure probably contributed to ROS formation and lead to the higher 8-OHdG level. The higher pH also induced the higher 8-OHdG formation.
