A simple, rapid, and sensitive spectrophotometric method has been developed for the quantitative determination of metoclopramide hydrochloride (MCP) in both its pure form and pharmaceutical formulations. The method is based on converting the metoclopramide hydrochloride drug to diazonium salt by using sodium nitrite and hydrochloride acid, followed by coupling with thiamine (vitamin B1) in a basic medium to form a pinkish-red colored species. Which showed maximum absorption at λmax 501 nm.By considering MCP as the analyte, the optimum conditions were studied and the method was shown the following parameters, Beer's law applied the concentrations ranging from 10 to 55 µg / mL, with a molar absorptivity of 8.843x103 L.mol−1.cm−1. The limit of detection (LOD), and limit of quantification (LOQ) are 0.801 µg / mL and 2.67 µg / mL respectively. Sandell’s sensitivity is 0.074 µg.cm-2, with recoveries ranging from 99.5 to 95.6 percent. The method has been successfully applied for the determination of metoclopramide in its pharmaceutical preparations.
