Capsules made from water-soluble chitosan (WSC) have been synthesized by adding of alginate. Capsules were made with variation of WSC:Alginate, i.e. 2:1, 3:1, 4:1, 5:1 and 6:1 (w/w). Chitosan used in this research was commercial chitosan from crab shell. Chitosan was depolymerized into water-soluble chitosan by hydrolysis method with H2O2. Characterizations were performed by FTIR test, weight uniformity test, swelling test, degradation test, solubility test and drug release test. FTIR spectra showed that the obtained capsule had vibration from its constituent molecules, i.e. chitosan and alginate. The results of the weight uniformity test and solubility test indicate that the capsules were obtained eligible the requirements by the Indonesian Pharmacopeia. The best of swelling degree in water and 0.1 N HCl solution was 84.45% and 262.23% respectively. The best of degradation percentage in water and 0.1 N HCl solution was 97.28% and 59.52% respectively. The best of drug release tests in water and 0.1 N HCl solution were 40 seconds and 1 minute 6 seconds respectively. Capsules that have the best physical appearance or texture and close to commercial capsules were WSC:Alginate 2:1.
